# Antibiotic intermediates (6-APA, 7-ACA): Overview

The two cores behind penicillins and cephalosporins

Source page: https://www.materialinsights.org/chemicals/antibiotic_intermediates/overview/

What this tab covers What the chemical is, how big its market is, what it is used for (with shares, volumes, why it wins and what could replace it), and its chain from raw material to end user. Ends with three things to remember.

Beta-lactam antibiotics such as amoxicillin, ampicillin and ceftriaxone are built on two cores. 6-APA is cut from fermented penicillin G with an enzyme; 7-ACA is cut from fermented cephalosporin C. Both need huge fermenters, cheap sugar and cheap power, and China dominates both. About 87% of India's antibiotic imports come from China. Aurobindo's Lyfius plant at Kakinada, with 15,000 t/yr of penicillin G, restarted Indian production in 2024 under PLI, and in 2026 India set minimum import prices and opened anti-dumping probes to protect it.
Volume not published globally; India's new capacity: penicillin G 15,000 t/yr (Lyfius), clavulanic acid 400 t/yr (Kinvan) Market value not published (market research figures for 6-APA are paywalled) Growth follows amoxicillin and cephalosporin volumes, about 3-5% a year (indicative) PSU Watch (2026); ORF (Dec 2025); growth indicative

#### Key uses

| Use | Share | Annual volume | Why this commodity | Alternatives |
|---|---|---|---|---|
| **Amoxicillin (via 6-APA)** | n/a | not publishedbasis | Amoxicillin is the most prescribed antibiotic worldwide and the largest outlet for 6-APA. It is made by coupling 6-APA with a D-hydroxyphenylglycine side chain, now mostly by enzymes. stoichiometry; qualitative | Cephalosporins Macrolides |
| **Ampicillin, cloxacillin and other semi-synthetic penicillins (via 6-APA)** | n/a | not publishedbasis | Different side chains on the same 6-APA core give different activity and stability. qualitative | Amoxicillin |
| **Cephalosporins: ceftriaxone, cefazolin, cefuroxime (via 7-ACA)** | n/a | not publishedbasis | Injectable and oral cephalosporins are hospital mainstays. India is a large ceftriaxone formulator and imports most 7-ACA from China. stoichiometry; qualitative | Carbapenems Penicillins |
| **Oral cephalosporins via 7-ADCA (cephalexin, cefadroxil)** | n/a | not publishedbasis | 7-ADCA can be made directly by engineered Penicillium fermentation, a route pioneered by DSM (now Centrient). qualitative | Amoxicillin |
| **Penicillin G and V as APIs and veterinary use** | n/a | not publishedbasis | Benzylpenicillin injections remain essential for syphilis and some infections; large veterinary use. qualitative | Other antibiotics |

No public source splits 6-APA and 7-ACA demand by downstream antibiotic. Shares left null.

#### From raw material to user

1. **Sugar or starch feed** Glucose from maize starch and soybean or corn steep liquor feed the fermenters; Lyfius has its own automated starch and glucose plant (Aurobindo, 2024).
2. **Fermentation** High-yield Penicillium chrysogenum strains make penicillin G; Acremonium chrysogenum makes cephalosporin C, in fermenters of 100-300 m3 (indicative).
3. **Recovery** Broth is filtered, extracted with solvent (butyl acetate) and crystallised as penicillin G potassium salt.
4. **Enzymatic cleavage** Immobilised penicillin G acylase removes the side chain to give 6-APA; about 1.7 kg penicillin G potassium per kg 6-APA at 100% yield (stoichiometric). Two enzymes convert cephalosporin C to 7-ACA.
5. **Semi-synthesis** Side chains are coupled to make amoxicillin, ampicillin, ceftriaxone and others, in dedicated beta-lactam facilities.

#### Three things to know

- About 87% of India's antibiotic imports come from China (Trade Map via ORF, Dec 2025).
- Aurobindo's Lyfius plant at Kakinada has 15,000 t/yr of penicillin G capacity and cost about Rs 2,270-2,500 crore (PSU Watch 2026; Aurobindo 2024).
- India's 2026 minimum import prices are Rs 2,216/kg for penicillin G potassium, Rs 3,405/kg for 6-APA and Rs 2,733/kg for amoxicillin (Medical Dialogues, Jan 2026).
