Antibiotic intermediates (6-APA, 7-ACA): Production route

How moulds and enzymes make the core of antibiotics

What this tab covers How it is made: the main production routes or, for product families, the main product types and technologies, compared on share of supply, inputs per tonne, plant scale, yield, co-products and unit cost.

Every beta-lactam core starts in a fermenter. Penicillin G is fermented, recovered as the potassium salt and split enzymatically to 6-APA. Cephalosporin C is fermented and converted to 7-ACA by two enzymes (D-amino acid oxidase and glutaryl acylase) or in one step with engineered acylases. 7-ADCA can be fermented directly. Enzymatic routes replaced older chemical cleavage using silylation and chlorinated solvents at very low temperature, cutting solvent use. Scale, strain productivity and power cost decide who wins.

How each route works

Penicillin G fermentation

Fed-batch aerobic fermentation of Penicillium chrysogenum with glucose and phenylacetic acid as side-chain precursor, over about a week. Broth is filtered, acidified, solvent-extracted and crystallised.

very scale-sensitive; needs cheap power

Aurobindo (2024); Medical Dialogues (2026); World Trade Scanner on DGTR (2026) Enzymatic 6-APA

Penicillin G potassium is hydrolysed in water by immobilised penicillin G acylase at mild pH and temperature; 6-APA is crystallised and phenylacetic acid recovered.

clean aqueous process; tied to penicillin G cost

Medical Dialogues (2026); stoichiometry Cephalosporin C fermentation and enzymatic 7-ACA

Acremonium chrysogenum ferments cephalosporin C; enzymes remove the aminoadipyl side chain to give 7-ACA.

India has no confirmed commercial 7-ACA plant (not confirmed)

stoichiometry Clavulanic acid fermentation

Streptomyces clavuligerus ferments clavulanic acid, a beta-lactamase inhibitor combined with amoxicillin (co-amoxiclav).

unstable molecule; specialised handling

PSU Watch (2026) Unit economics are indicative ranges from the sources named; costs move with feedstock and energy prices, plant size and region.
