Small-molecule APIs: Overview
The active molecules inside every pill, and who makes them
What this tab covers What the chemical is, how big its market is, what it is used for (with shares, volumes, why it wins and what could replace it), and its chain from raw material to end user. Ends with three things to remember.A small-molecule API is the active chemical in a tablet, capsule or injection, made by multi-step organic synthesis or fermentation and purified to pharmacopoeia standards. The merchant and captive API market is worth roughly $200-250 billion a year (indicative). India is the third-largest medicine producer by volume and makes about a fifth of the world's generics, but in 2023-24 it imported Rs 37,722 crore of bulk drugs and intermediates, more than 70% from China. That dependence, and the PLI scheme built to cut it, is the core story for an Indian investor.
Volume not published as a global tonnage; individual APIs range from a few kilograms a year (oncology, peptides) to over 100 kt (paracetamol, metformin) Market value about $200-250 billion (2024-25, indicative, basis: market research estimates vary widely; includes captive API made by originators) Growth about 5-7% a year to 2030 (indicative, market research consensus); generic API prices deflate while volumes grow Department of Pharmaceuticals Annual Report 2024-25 via ORF (Dec 2025); Pharmabiz synthesis of official data via ORF; market size indicativeKey uses
| Use | Share | Annual volume | Why this commodity | Alternatives |
|---|---|---|---|---|
| Generic oral solid medicines (tablets, capsules) | n/a | not publishedbasis | Most API tonnage goes into off-patent tablets: analgesics, antidiabetics (metformin), cardiovascular drugs (sartans, statins), anti-infectives. Price per kg is low and cost leadership decides who wins, which favours large Indian and Chinese plants with backward integration into KSMs. qualitative; ORF (Dec 2025) | None for the molecule; competition is between suppliers and routes |
| Anti-infectives (antibiotics, antivirals, antiretrovirals) | n/a | not publishedbasis | Beta-lactam antibiotics rest on fermentation intermediates (penicillin G, 6-APA, 7-ACA) where China is dominant. Antiretrovirals for Africa and global health tenders are a large Indian API niche (Laurus, Aurobindo, Hetero). ORF (Dec 2025); Laurus Labs FY26 results | Vaccines and non-antibiotic therapies reduce some demand |
| Patented and innovator small molecules (captive or CDMO-made) | n/a | small tonnage, high valuebasis | Originators make some API in house and outsource the rest to CDMOs such as Lonza, WuXi AppTec, Divi's and Siegfried. Value per kg can be thousands of dollars, so quality and IP security count more than cost. Lonza FY2025 results; GEN Top 10 CDMOs 2026 | Biologics and peptides displacing small molecules in some areas |
| Veterinary and animal health | n/a | not publishedbasis | Shares many generic APIs and antibiotics with human medicine. Laurus lists animal health among its FY27-28 capex areas. Laurus Labs FY26 press release | Vaccines, feed additives |
From raw material to user
- Basic chemicals and solvents Benzene, toluene, phenol, methanol, acetic anhydride and chlorinated solvents from refineries and chemical plants; India imported about $26 billion of HS 29 organic chemicals in 2024 (Trade Map via ORF).
- Key starting materials (KSMs) First registered building block in the regulatory filing; China supplied about 70% of India's critical bulk drug inputs (Department of Pharmaceuticals 2020-21).
- Intermediates Several synthesis steps under GMP; a typical small molecule uses 168-308 kg of materials per kg of API (process mass intensity, ACS GCI Pharmaceutical Roundtable via Kekessie et al. 2024, indicative).
- API synthesis and purification Final step, crystallisation, drying and milling to a pharmacopoeia grade; registered with US DMFs, EU CEPs and CDSCO.
- Formulation Mixed with excipients into tablets or injectables; India exported about $30 billion of medicines in FY25 (Pharmexcil, indicative).

Oil Crude oilThree things to know
- India imported Rs 37,722 crore of bulk drugs and intermediates in 2023-24, up from Rs 24,172 crore in 2019-20, and more than 70% came from China (official data via ORF, 2025).
- The bulk drug PLI scheme (Rs 6,940 crore outlay) had drawn Rs 5,210 crore of investment and Rs 3,792 crore of sales by June 2026, small against the import bill (PSU Watch, 2026).
- In 2019 only 28% of facilities making APIs for the US market were in the US, against 26% in the EU, 18% in India and 13% in China (US FDA testimony, Oct 2019).
