Pharma starting materials and intermediates: Overview

The building blocks behind paracetamol, metformin and antibiotics

What this tab covers What the chemical is, how big its market is, what it is used for (with shares, volumes, why it wins and what could replace it), and its chain from raw material to end user. Ends with three things to remember.

A key starting material (KSM) is the first registered building block in an API's regulatory dossier; drug intermediates are the compounds made between it and the final API. They are mostly heterocycles, nitro and amino aromatics, and fermentation products, made in chemical plants rather than GMP suites. India's Department of Pharmaceuticals found China supplied about 70% of its critical bulk drug inputs, and the 2020 PLI scheme targets 41 such products. Para-aminophenol for paracetamol, dicyandiamide for metformin and penicillin G for antibiotics are the best-known cases.

Volume not published globally Market value not published as a separate market; included within India's Rs 37,722 crore (about $4.5 billion) of bulk drug and intermediates imports in 2023-24 Growth tracks generic API volumes, about 5-7% a year (indicative) Department of Pharmaceuticals Annual Report 2020-21 and official data via ORF (Dec 2025); growth indicative

Key uses

UseShareAnnual volumeWhy this commodityAlternatives
Fermentation-based KSMs (penicillin G, 7-ACA, erythromycin thiocyanate, clavulanic acid)n/a Lyfius penicillin G 15,000 t/yr capacitybasis Beta-lactam and macrolide antibiotics all start from fermentation products. Scale and cheap power matter most, and China's large plants closed most Indian fermentation capacity in the 2000s (indicative). PSU Watch (2026); Department of Pharmaceuticals PLI guidelines (2020) None at scale; synthetic routes are uneconomic
Chemical-synthesis KSMs for high-volume generics (para-aminophenol, dicyandiamide, 2-methyl-5-nitroimidazole, 1,1-cyclohexanediacetic acid)n/a Meghmani para-aminophenol 13,500 t/yrbasis These feed paracetamol, metformin, metronidazole and gabapentin, some of India's largest-volume APIs. They are bulk chemicals made by nitration, hydrogenation and condensation. PSU Watch (2026); PLI guidelines (2020) Alternative routes from different KSMs
Heterocyclic intermediates for sartans, statins, antiviralsn/a not publishedbasis Multi-step building blocks (tetrazoles, biphenyls, pyrimidines) where China and India both make large volumes. Centrient's atorvastatin plant (206 t/yr) is a PLI example. PSU Watch (2026) In-house synthesis by API makers
Advanced intermediates for innovator APIs (CDMO)n/a not publishedbasis Late-stage intermediates made under GMP for originators. Divi's moved pre-chemistry raw materials to its Kakinada Unit 3 in Q4 FY26 for backward integration. BoB Capital on Divi's (May 2026) China (WuXi STA, Asymchem)
No public source splits KSM and intermediate demand by product. The PLI scheme groups 41 products into four categories: fermentation-based KSMs and drug intermediates, fermentation-based niche KSMs and APIs, chemical-synthesis KSMs and drug intermediates, and other chemical-synthesis APIs (Department of Pharmaceuticals guidelines, 2020).

From raw material to user

  1. Basic chemicals Benzene, phenol, nitrobenzene, ammonia, acetic anhydride, cyanamide; organic chemical imports from China rose from about $0.29 billion (2001) to over $11.1 billion (2024) (Trade Map via ORF).
  2. Simple intermediates Nitration, chlorination and hydrogenation make nitro- and amino-aromatics such as p-nitrophenol and p-aminophenol.
  3. Key starting material The registered KSM fixes the point from which GMP and regulatory change control apply.
  4. Drug intermediates Further steps under GMP lead to the API; Divi's and Laurus integrate several steps back.
  5. API See apis tile.

Three things to know

  • China supplied about 70% of India's critical bulk drug inputs (Department of Pharmaceuticals Annual Report 2020-21).
  • The PLI bulk drugs scheme had 48 projects approved and 38 commissioned by December 2025, with about 56,800 t/yr of capacity (IndraStra citing PIB, 2026).
  • Imports averted by PLI were only about Rs 2,192 crore cumulatively by December 2025, against annual bulk drug imports of Rs 37,722 crore (IndraStra; ORF).
