Pharma starting materials and intermediates: Production route

Nitration, condensation or fermentation: how KSMs are made

What this tab covers How it is made: the main production routes or, for product families, the main product types and technologies, compared on share of supply, inputs per tonne, plant scale, yield, co-products and unit cost.

KSMs are made either by bulk organic chemistry or by fermentation. Chemical KSMs come from aromatic feedstocks through nitration, reduction, chlorination and condensation, often in plants that also make dyes or agrochemical intermediates. Fermentation KSMs need large aerobic fermenters, cheap glucose and cheap power. The economics are those of fine chemicals: batch plants of a few thousand tonnes, where solvent recovery and effluent handling decide cost.

How each route works

Nitration and hydrogenation (amino-aromatics, e.g. para-aminophenol)

Nitrobenzene is hydrogenated in sulphuric acid over a platinum or palladium catalyst and rearranges to para-aminophenol (Bamberger route), or phenol is nitrated to p-nitrophenol and reduced. Product is crystallised and dried.

cheap at scale; acid effluent

PSU Watch (2026); stoichiometry Condensation and heterocycle synthesis

Cyanamide, amines and carbonyls are condensed into imidazoles, pyrimidines, biguanides and tetrazoles. Dicyandiamide (for metformin) comes from calcium cyanamide.

commodity-like; China dominant

qualitative Fermentation KSMs

Penicillium or Acremonium strains produce penicillin G or cephalosporin C, which are extracted and converted to 6-APA and 7-ACA. See antibiotic_intermediates.

capital-intensive, scale-dependent

Chlorinated and fluorinated aromatics made from chlorine, HF and aromatics; feeds many APIs and agrochemicals.
